The fact that DNA cannot be used for identification in some cases due to the small amount of sample or degradation due to environmental conditions increases the importance of RNA. In this direction, studies are carried out on the use of miRNA markers in the detection of body fluids in forensic sciences. The focus of the study is the identification of the miRNA biomarkers hsa-miR-203a-3p and hsa-miR-451a, especially in bloodstains exposed to environmental conditions. The samples that were created by the modeling method were kept in different environmental conditions for 45 days and then miRNA markers were investigated. As a result of the analysis, hsa-miR-451a expression was observed in the samples kept at -20°C and +4°C and in sealed bags, while hsa-miR-203a-3p expression was not observed. This study proposes the use of RNA and especially miRNA instead of DNA in forensic sciences. In conclusion, this study points to the importance of using miRNA profiling in forensic science. In particular, the identification of bloodstains taken from the crime scene and exposed to various environmental conditions with the miRNA biomarker hsa-miR-451a may be an important step in future forensic science applications. Since this study was carried out as a master's thesis, it was studied with a small sample group, and the results were intended to form a basis for future studies. Increasing diversity with larger sample groups in future studies is scientifically valuable.
