MYCOBACTERIUM TUBERCULOSIS 76-KDA ENOYL REDUCTASE ANTIGEN PURIFICATION AND CHARACTERIZATION

Abstract

Unlike most other bacteria, Mycobacteria make fatty acids with the multi-domain enzyme eukaryote-like fatty acid synthase I (FASI). This study aims to clone and express 76-KDa enoyl reductase antigen from Mycobacterium tuberculosis. This antigen was expressed and found in an insoluble form as recombinant protein in Escherichia coli. The results showed that the enoyl reductase domain from FAS1 is composed of 2121 bp and it encodes for a 76 KDa proteinas expressed in E. coli may be useful for detecting human IgA- and IgG antibodies against active TB. However, for more specific results, larger sample panels (healthy, other diseases, TB) need to be evaluated in ELISA and other rapid tests.
