The matK gene analysis in five types of matoa (Pometia pinnata J. R. Forst. and; G. Forst) based on the pericarp colour

Abstract

The diversity of matoa (Pometia pinnata, J. R. Forst. and G. Forst.) was grouped based on the morphology of the pericarp color there were green, red, yellow, brown, and purple. However, the DNA sequence of these cultivars is undetermined. This study aims to determine the variation of DNA nucleotides and the kinship of matoa cultivars based on matK genes. DNA extraction was performed using a genomic DNA mini kit. DNA amplification using matK-barP-390F and matK-barP-1326R’ primer sets. The sequence of nucleotides was aligned and compared using the basic local alignment search tool program of the National Center for Biotechnology Information. The phylogenic reconstruction was carried out using maximum likelihood and maximum parsimony. Based on this study, matoa has 905 bp–1286 bp with similarity values of 98.08%–99.40% on Pometia tomentosa AF314802.1. The genetic distance between the matoa in this study was 0.0000–0.0046. The phylogenetic tree shows that all five matoa were divided into two clusters; the first cluster included yellow, green, and red matoa, whereas the second cluster was brown and purple matoa. The results of this study validated the unility of matK genes in matoa identification, which can support plant breeding by providing valuable genetic information.
