Application of Fourier transform infrared (FTIR) spectroscopy coupled with multivariate calibration for quantitative analysis of curcuminoid in tablet dosage form

Abstract

Rapid and reliable method based on FTIR spectroscopy in mid infrared region in combination with multivariate analysis has been developed for prediction of curcuminoid content in tablet formulation. In this study, FTIR spectra was subjected to several optimization including wavenumbers selection and derivatization to get prediction models between actual values of curcuminoid as determined using reference method of high performance liquid chromatography and calculated values. The first derivative FTIR spectra at wavenumbers of 2975-660 cm-1 using partial least square (PLS) calibration was preferred for quantification of curcumin in tablet with R2 in calibration and validation of > 0.99, while desmethoxycurcumin was predicted using PLS regression exploiting normal FTIR spectra at wavenumbers of 1784-1587 cm-1 with 5 factors with R2 values for calibration and validation models of 0.9954 and 0.9916, respectively. FTIR spectroscopy combined with PLS regression can be used a rapid analytical method for determination of curcuminoid in tablet dosage form.
